Homology Modelling

Homology modelling, also called comparative modelling, is a method used to predict the three-dimensional structure of a protein from the known structure of a similar protein.

The method is based on the idea that proteins with similar sequences can have similar three-dimensional structures.

Main Steps

  1. Template identification: find a similar protein with a known structure.
  2. Sequence alignment: compare the target protein sequence with the template.
  3. Model generation: build a three-dimensional structure using the alignment.
  4. Model refinement: improve the geometry and structure of the model.
  5. Quality assessment: evaluate the quality and reliability of the predicted structure.

Homology modelling is especially useful when the experimental structure of a protein is not available. It is widely used in structural biology, bioinformatics, and biomedical research.
